Yeah, I’m with A304, Big Jim, Cathy, Josh Harber and some of the other staff confirmed that that was their relationship, a brother/sister one, and as for the whole parent thing, think of them as Nani and Lilo.

  1. I don’t care about fan interpretation. Some said he was a slave, glad this was proven false.
     
  2. People have wanted them to say something forever, and now he’s confirmed to be her foster brother, and their relationship pretty much reflects an older sister and younger brother, he obviously gravitated more to Twilight than Shinning and half of his life was raised by her after he left home with her. Think Lilo and Nani.
     
    Big Jim confirmed that their relationship was like Lilo and Nani, some other people behind the show also said they saw their relationship like brother-sister, so their you go, fans can suck it.
     
    Also I’d like to add, Cathy had every right to do this, I’m sorry but the other interpretations, make me want to kill Twilight.

3 things here.  
  1. the episode is Sparkle’s seven.  
  2. Sparkle’s Seven was written by the voice actors, their personal bias is going to leak through in that regard, Cathy has always been of the mind that Spike was more of a brother to Twilight and Shining than anything else, and I personally think it was kind of a dick move on her part to try and cement their relationship like that, instead of leaving it open to fan interpretation.  
  3. the episode really does directly contradict the implications of Father know’s beast, which came before it, and held some heavy hints that Spike saw Twilight in a parental light. Recall his choice of words during one of their arguments in the episode  
    “Maybe you don’t like it now that I have a real parent!”  
    Even if it was a heat of the moment thing, it implies that he did see her as a parental figure in someway, though the writers did a good job of leaving it open to interpretation by having Spike confirm to Twilight that he thought of her as family, with no specific familial role mentioned.
