First: Its a animation style to show Spike as a more important character than Winona.
 
He have more unique design with lizard eyes, standing out like something not from this world. It was original meant just for Spike transformation from a dragon but they keep it post Rainbow Rocks.
 
And about Winona - she is just a dog and her eyes look like dog eyes IMO.
