Somehow I imagine her being a chemistry teacher (along with Zecora, perhaps) that has quite a few problems with school administration due to her experiments and demonstrations being a little too spectacular for everyone’s safety. But students always stand up for her. And they don’t mind being called “my little minions”. Not at all.
That whole brief annoyance followed by smiling thing is common to both, also she could have just been in the library helping, teachers tend to have allot of range in what their jobs are,
The EQG one seemed a lot less… well, cheery. Also, she appears to be both a teacher and a librarian, due to reusing her character model.
What was different about them?

Well, the Mane-iac is fictional to ponies, so maybe that’s just a cosplayer.
uotapo’s humanizations are great but sometimes ooc imo.

No, I’d say she either teaches biology or chemistry. Which actually works very well with my headcanon that most of the science teachers are villains.
