Okay, I’ll give you my honest opinion on this.  
You haven’t put as much work into Big Mac, and the random human as you actually put into Pinkie Pie.
 
Background is OK but could use some work. Don’t make it a simple floor and wall type thing. Be more creative and think what type of things she would have in this room. Cutie mark may be a little out of place as she is anthro/human, but that’s just my opinion.
 
Seems like you may have gotten a little lazy with the background, and the human and Big Mac.
 
Other then this, the picture is pretty good.

Might I ask if you upload just to derpibooru, or do you have a deviantart account or something of the like, just curious.  
Not sure on how good I am with art critique, but I will say that this is pretty good for your first image uploaded here. I’d be happy to see more of this.

Seems like you have problem with shadows.  
I suggest that instead of setting the opacity of the brush, You start a new layer just for the shadows, And set it’s opacity instead, This way it won’t overlay itself, And you could make more defined shadows.  
Also don’t afraid to use layers, That’s probably the best advice, Honestly sometimes i even use 30+ layers on a simple drawing.  
Ah and always start with a huge canvas, So you have more options for brushes and work more freely, And later on you can re-size it. (i really hope you don’t actually work with 900x500 canvas).
 
But overall it looks pretty decent, You obviously know what you’re doing

You have a good sense for hands/feet-details, but give also the background a bit more of attention, it really adds a lot to the atmosphere.

kay dude first of all download paint tool.sai or photoshop dont use ms paint
