This is hard. I love all of them (but none in a romantic/waifu sort of way). So confessing the love and doing bad things to them is not good.
 
I’d punch AJ on the elbow. She seems tough and strong. I won’t hit her all too hard though.
 
I’d confess my love to Rarity. She’s classy. Even though I’m not into other girls, I bet a date with her would be fun.
 
Tickle the crap out of Rainbow Dash. …With her own feathers.
 
I’d roast Pinkie Pie. She has a good enough sense of humor I think to not mind a bit of ribbing at her own expense. Also, she’s my least favorite of the six. So even though I love her as a character, I do think I have enough ammo to throw at her.
 
I’d play a video game with Twilight. She might like that.
 
And I’d ignore Fluttershy. She is an introvert and shy so I don’t think she’d mind.

Join a fight club with Applejack (She may be stronger than I am, but that doesn’t mean I can put up a fight)  
Write a book with Twilight (Do I even need to explain this one?)  
Bake something with Fluttershy (It’s more relaxing than the other choices in my book)  
Join a tabletop gaming group with Rarity (As long as she doesn’t turn it into a fashion show)  
And a date night with Pinkie Pie (because she’s Best Pony!)

I’d roast Rarity (like a marshmallow)  
Punch Applejack (She isn’t one of my faves anyway)  
Ignore Fluttershy (She’s too quiet to listen to, anyway)

1: Says Mane Six, but it doesn’t state they have to be from the same dimension, so the Rarity I knocked out was from DRESS.MOV
 
2: Which kind of roast? If by roast meaning cooking, then Cupcakes Pinkie Pie is dinner.

Hm… Punch goes to Rarity, since a gala without punch is worthless. Roast goes to AJ, she has the best backyard. I’d ignore Pinkie Pie for all of three hours while my headache recedes. I’d tickle RD, just because I can. I’d play videogames with Twilight because my friends all suck at Portal and I’d confess my love to Fluttershy, simply because she’s my type. Your move, artist.

Alright, let’s do this:

Oh goody, it’s tough decision time again, isn’t it? Alright, let’s see:
 
Roast Applejack, as she would probably take it better than the others.  
Tickle Pinkie, because she seems to be the type who would enjoy it most.  
Play a Videogame with Twilight. I was initially considering Rainbow Dash for this one, but then I remembered Rainbow Rocks.  
Confess my love to Rainbow, on the grounds that she is my favourite of the Mane Six.
 
That leaves only Rarity and Fluttershy, to be punched or ignored respectively.
 
I don’t think I’ll be punching Fluttershy, as she probably wouldn’t tke it too well. So I suppose fluttershy will be getting ignored.
 
So Rarity get’s punched, on the grounds that she’s the only one left.

This is hard but if I had to make a choice
 
Rarity’s fabulous, but she’d be the easiest to ignore  
Twilight would probably know to ignore insults so i’d roast her  
Fluttershy would be the one i’d tickle the crap out of because y’know, Who Wouldn’t  
Pinkie would be the most fun to play a video game with so i’d probably go with that  
Dash is my fav so i’d confess my love to her  
AJ… i regret trying to make decisions, sorry AJ
