It was already confirmed in Twilight’s Kingdom when she transferred her magic to Twilight and her mark faded away.
I guess this also confirms that the black splotch on her flank is just a birthmark rather than part of her cutie mark.
Also no cutie mark on Luna? According to the Journal of the Two Sisters, Luna and Celestia were still blank flanks despite being fully grown due to Alicorn’s having different life cycles. Which means this series must be set during that time period where Starswirl was still mentoring them to become the new Princesses of the recently founded Equestria.
