It doesn’t really matter for Twilight from the series. In season 5 finale, filly Rainbow Dash says to her, “I mean, you’re a full-grown pony, and… Wait. Are you an Alicorn?” What means Twilight is an adult.

Faust claimed the pony mane 6 are young adults (18-22) and the mane 6 in EQG are (17-18 senior) while the CMC in EQG are (14-15 freshman) so aging probably works differently there than in the pony verse it’s safe to assume

I asked my own watchers on DeviantArt for theories about this and one had a theory that it might be because ponies have naturally longer lifespans than humans, thus making their ages more spaced out. That makes enough sense in my eyes. It’s also why Celestia and Luna are still alive there since there wasn’t any magic before Sunset came.

I dunno, if their ages are the same as in the EG universe, technically these two are both teenagers.  
Flash is from the EG world and there’s not much age difference between Twi and him from what we can tell.  
(I also recall Faust mentioning the pones being 19/20 at most) I might be wrong tho. XD
 
Until we get official ages, which we probably won’t, we’ll never really know!
