Cadance is the Princess of Love. Love makes you do crazy-ass things for the sake of the one(s) you care about. She’s also the only one on this list who has killed in order to protect others, not to mention the only one on the list confirmed to have killed at all. (Okay, so she didn’t kill Sombra on her own, but she certainly led the charge, so it seems fair to say she had lethal intent.) I think that counts as “stepping up to do something you don’t want to do.”

“I’d die for you first” is the emo version of “No, I love you more.”

I read it as such:  
“I’d die for you” is a metaphor for “I’d do things for you, you don’t want to do.”  
And the reactions of the ponies is appropriately scaled down:  
“Then perish” is basically “Yes, I expect you to.”  
“You will.” is more or less “I know, I manipulated you to.”  
“Please don’t.” is just that. Apparently with different thoughts behind it. (Ie. “I’ll do it myself” in AJs case.)  
“Cool.” is probably literally their reaction to it.  
“I’d die for you first.” has to be scaled down to fit the situation but basically means “Before I let you do something you don’t like to do, I’ll do things you don’t like to do.”
 
As for the chosen alignments, I don’t actually agree with all of them. Rarity is more “You will”, Sweetie Belle feels more like “Cool.”, I’d prefer to see AJ in “I’d die for you first.” and I am really not sure about Cadence at all.

Which is exactly why I feel the first two lines should be practically empty, none of them would say such mean things really.
