This seems out of character. Pinkie is an ambiguously-employed baked-goods shop gofer who parties nonstop. A coke habit is way out of her league, financially. She probably uses meth. Twilight Sparkle is the rich Canterlot native who’s constantly on edge, who might have a coke habit.
 
Rarity probably abuses Fluttershy’s diet pills, to help her stay awake and get through her larger dressmaking orders on time (and also keep her figure, not that the pills are needed for that). Fluttershy for her part uses her diet pills exactly as prescribed. However, she often spends long periods in the wilderness with her animal friends, and the more colorful mushroom species.
 
No shock, Rainbow Dash might be doping, though she denies it. In any case, it wasn’t her idea. Spitfire basically laid it out that being a Wonder bolt takes a pony’s all, and skipping the blood doping, erythropoeitin, furosemide, and anabolic, wasn’t somepony’s “all.”
 
And Applejack, not even a secret. Those apples that started to ferment have to go somewhere. Maybe they don’t actually have to be fermented more, pressed, and distilled to quite so much proof, but it’s not like they can take her legs away for another GWI (galloping while intoxicated) arrest.
