My guess is that this universe does have a Twilight but judging by that image my guess is that the whole Nightmare Moon incident never happened so she never had to “make some friends”, so this worlds Twi stayed a unsocial bookworm who only threw herself into her studies.
 
As for where she is now? My guess is that shes in Equestria, that when (pony) Twi was sent to the human world (human) Twi was sent to Equestria.
 
So we might get some scenes were we cut back to Equestria to see how (human) Twi and (pony) Twi’s friends are doing as they help (human) Twi along and teach her the Magic of Friendship, so when she returns to her own world she can become friends with (pony) Twi’s friends’s counterparts (so long as they have the same personality).

Snips and Snails are both ponies, the same species as Twi and the rest.
 
Spike is a dragon, that’s why he isn’t a human but something else.
 
The human world seems more mundane, so humans are probably the only known sentient species.
 
The alternative would probably have been to turn Spikei into a gecko or something, which wouldn’t settle as well with the kiddies compared to an adorable puppy.

Given the push of Alicorn Twilight toys that they are making for this new season, and that Meghan said that SS wouldn’t be on the show (this, technically, is not), I’m not worried this is TS’s swan song.

Bu- Bu- But the laser and orbital strike cannon which probably is the final battle during the last 10 minutes of the movie? Im not sure how TS will change SS in the last few minutes of the movie (unless they do some lazy writing like Trixie and other villain redemptions).
 
Also, do you honestly trust Tara Strong’s words for a 100%? Maybe when she meant: “TS is staying in Ponyville” she meant HUMAN ponyville. And it’s PR, you’ll tell anything good to not scare the fans away.

More than likely, Twilight “grooms” SS into being the 6th for the human 5 before returning.

-Her story is kinda complete in FiM (princess and all). And im more likely to believe that the writers want to keep FiM’s S4 in that ‘status quo’ area (the fish out of water, needs to make friends kind of character, which SS could be)  
-I wouldnt be surprised if Twilight at the end believes she needs to stay to help her human friends.  
-That group high five at the end of the trailer might hint that: yeah Twilight will be staying here with her new friends.
