Spike’s reaction is fair, for he is doing is job and his thought process when it comes to parents is different. An orphan would love to know his parents, and they wouldn’t quite like someone else who they view as “lucky” enough to have them disliking their parents. Of course, he’s a fool because parents can actually be very unpleasant people or overbearing on their child to a point where it wears on them immensely, but I can understand him.  
Rainbow, on the other hand (I just noticed that whoever posted this here forgot to add the tag for Rainbow Dash), is just being a moral busy-body asshole who views someone thinking that their parents hate them as them thinking the whole bloody world revolves around them. She’s got completely and utterly broken logic. Now, of course, this AU’s Twilight does have an ego, but her saying what she said makes no bloody sense at all. It’s a complete non-sequitur, she’s taking two things which, as far as we know, are completely unrelated, and trying to use it to… be an asshole. It would be like if someone was, say, a thief, and one day a branch breaks off a tree on his lawn, and he doesn’t clean up the branch, and then someone else got extremely upset at this, and a third person says, “I hope this person being upset at this branch has taught you that you shouldn’t be a thief!” Now, of course, the analogy is only supposed to work when applied to Rainbow. It’s completely flawed when applied to Spike’s issue with this, but it’s not meant to be applied to that.
