Fluttershy is an expert in animal biology and basic veterinary science, and perhaps environmental engineering depending on how much experience she’s got in Fluttershy Leans In.
Applejack knows all about botany and farm economics, with good logistics and agricultural management skills. She always insists appuls are healthy for ponies, so she might be into nutrition in some ways.
Pinkie is BORN to party and to bring joy, which means positive psychology and emotion management are her gifts. Combining Applejack’s cooking, they’d present us excellent culinary art classes. Pinkie teaching diplomacy is an interesting idea, but she probably knows less than Twilight, politically speaking.
Rarity easily nails business management. Just look at how successful her boutiques are! Art classes, or introductions to designing, handiwork, art history, and even theater arts, but I’m not sure the way fashion is taught in institutions.
RD is the physical education coach there, but always be reminded that she’s led a hurricane team, showing her in-depth knowledge to atmospheric sciences and meteorology.
Twilight is the know-it-all among the six. She was even able to give a lecture on pegasus physiology/physics in Top Bolt.
Spike can give much info on the dragon species (though the lore tells little to us); Starlight is an extremely powerful sorcerer, so she can cover advanced magic courses, and, well, from her past experience, an introduction to authoritarianism and the knowledge on cutie marks. Maud’s got a Rockology Ph.D. Cheerilee is a good educator of elementary level. Big Mac knows more about logistics and agri management. The CMC, self-inspiration. Discord, individual study time. The Mayor Mare, basic political science.
And there are the royalties. I seriously have no idea what they can cover there.

A pillar teaching history reminds me of the episode of Supermansion where American Ranger tries to teach American history but never gets around to it because after skimming through the textbook and learning about the terrible things that happened because of/to various presidents, reacts poorly.
Worst case for Starswirl would be a few awkward moments interacting with Luna trying to bring up the whole Nightmare Moon and banishment thing. That or Discord and his ups and downs and downs..and sideways because this is Discord.
That makes it all the better.

Well, Fluttershy could cover animal care and basic biology. Depending on how one interprets the nature of her job, she might also take a stab at veterinary medicine. Ecology, perhaps?
Applejack can easily handle agriculture, and given the requirements of running a farm also home management and likely some form of economy as well.
Pinkie’s a bit trickier, but besides baking and/or home ec, her interactions with the yaks and griffons make me think she could be a solid diplomacy instructor in a pinch.
Rarity can certainly handle economics – she is a professional businesswoman.
Rainbow… eh, she’s tricky. Gym? I can’t really think of any intellectual pursuit she would care very much about. Perhaps the making and management of weather?
Twilight could cover a lot of things, but I think she’d really shine in teaching magic and history.
Depending on who’s willing to be a guest instructor, Maud can probably be one of the best geology teachers in Equestria, if not necessarily the most riveting; any of the Pillars could provide an excellent insight into their specific culture in their time period insofar as history goes, and I don’t imagine Meadowbrook, Mistmane or Starswirl would mind giving a lecture or two about their fields of magic; Zecora could likewise have a lot to teach about alchemy and potion-making, as well as her own culture and the nature and wildlife of the Everfree.
