Both cases use the dual meaning of eight to mean ate, consume. In this case Derpy told the joke wrong, by starting on the wrong numbers. Saying six, seven eight.
No normal pony laughed because Derpy told the joke wrong and to them it doesnt make sense.
However. Six, seven eight is still a grammatically correct sentence. Just arranged unusually. To change it for ease of comprehension it becomes “As for Six, it was consumed by Seven”.
Twilight laughs because she recognised this instantly, and didnt realise until afterwards that the joke is being mistold.

The joke usually goes seven eigth(ate) nine, but derpy starts one number lower. Which still works actually, but in a weird grammatical way (Six, seven ate).
