Derpy started out as an animator in-joke that became public because Jayson Thiessen found it amusing; I don’t see anything like that happening in G5. There may be a silly wall-eyed pony popping up somewhere in the new series, but I wouldn’t expect too much out of her.

Yes, and her name was Surprise. However Pinkie Pie had been the franchise’s main “poster pony” for many, many years so Hasbro requested her be added to the lineup. So they took Surprise, removed her wings, added a reverse of Pinkie’s original color scheme (mane lighter pink than coat) and was done.
Pegasus Pinkie Pie was actually part of Lauren Faust’s initial ideas for FiM
and personally, I actually kinda like the idea of unicorn Fluttershy

Actually, they did make her a pegasus, but the emails indicated that in internal polls it was universally unpopular.
The concept art showed Fluttershy as a unicorn and Twilight as an Earth pony, though.

No, but everything is being changed. The only remaining things are the character’s names and likely their overall color schemes (not counting Applejack.) Not even their races are guaranteed to be the same. And so far no mention of any surviving characters outside of the Mane 6.

Yes.
We won’t, but Hasbro sure the hell will. The fact it’s a remake and the words “nuke it from orbit” about G4 coming to mind assures us they practically already have.
