No, they work in two totally different ways (the rainbow to ponyland is natural and takes you to an alternate realm [think difference between heaven, hell, purgatory and the real world, or Kingdom Hearts’ worlds, or the Fairy World and Dimmsdale in Fairly Odd Parents] the mirror is artificial, lost Crystal Pony technology made from magical crystals, and takes you to a semi-artificial mirror world [like Oz, Wonderland, or Neverland] and that the only thing the 5 generations [G1, G2, G3, G3.5, and G4] plus Tales have in common, is that they feature talking colorful ponies with symbols on their butts. If we do not include Tales, they all feature unicorns and pegasi, and have other fantasy creatures. Technically G3.5 is just G3 but nixed of all but 7 ponies, and with a completely changed design, and that’s it. But these are the only things they really share.

You seriously think the rainbow to Dream Valley works almost exactly like the magic mirror from Equestria Girls?  

I think that if TJ went on the rainbow into Ponyland, he would look the same (but talk) but he would not be very useful and (normal horse) + (ponyland pony) would only be able to produce the most unstable of offspring, if at all.
 
On the other hand, if a Ponyland Pony went to the human world, she would transform into an human herself, but still showing some pony traits; if Firefly went to the human world, she would still have blue hair, purple eyes, pink pony-like ears, pink pegasus wings, a blue pony-like tail, and double blue thunderbolt markings on her buttocks but also in her clothes. Gusty would retain her horn, Bubbles would retain her diamond facemark, Applejack\Bowtie would retain their freckles, the Seaponies would have cute little scales and fins, the Rainbow Ponies would still be glittery and have rainbow hair, ecc… And some ponies might even have variance in ethnicity!
