Her reaction to the mention of it, the reveal of its secret passage, her knowledge of forbidden secrets in EG1, and the semi canon tie in comics, all suggest she knew about this place already.
For this one scene, was she trying to act surprised for Twilight’s sake, or, more likely, the realization that Celestia was willingly showing her this room finally hit her? This was forbidden to even apprentices and other princesses until now.
Most likely the latter, as she was simply smiling as the secret passage opened up for them. She clearly knew about it.
