Twilight and Spike are definitely closer to being family than Cadance and Blueblood are (unless they’re both actually related, at which point it stops being “almost incest”. :P
Seriously, Twilight hatched him, and from what I can tell, raised him for at least part of his life. Their little family may be a bit dysfunctional (for example, those times when Spike was the one looking after Twilight instead of the other way around), but it’s gotten them this far.
She went to Celestia’s school for gifted unicorns, and her parents have been seen. It hasn’t been stated if Twilight lived in the castle with the princess or stayed with her parents.
