Well she has had more serious duties since defeating Tirek, since she’s gotten an official title, a castle and is expected to act more like a princess now compared to in Season 4 where it was mostly just her with wings.
Lack of sleep, lack of food…once Twilight Sparkle gets her mind set on something, she pursues it to the point of self-destruction. I’m pretty sure a big part of Spike’s job is keeping her from killing herself from overwork.
