The key difference is that Patachouli is a bit rude and dismissive of everyone else.

It really is. They have a LOT of similarities.
 
Both are extremely powerful in a wide variety of magic.
 
Both are librarians.
 
Both are shut-ins (hiding within their libraries).
 
Both are purple.
 
Both have assistants that they’ve “created”, and have a close bond with said assistant. (Twilight hatched Spike’s egg, Patchouli summoned Koakuma, a demon girl, from…somewhere.)
 
Both are often sarcastic as hell.
 
Both suffered as a result of their shut-in tendencies (Patchouli’s physical health, Twilight’s social skills)
